Evaluation of Bacterial Leakage of Root Canal Fillings over Time Using Modified Bacterial Leakage Model
Objective: To evaluate the bacterial leakage of root canal fillings over time using the modified bacterial leakage model.
Material and methods: One hundred and seventy mandibular premolars were divided into 6 experimental (n=25) and 2 control groups (n=10). 170 leakage apparatus were set up using Enterococcus faecalis as a microbial marker. The models were observed daily for turbidity of the broth in the middle and lower chambers for 7, 10, 15, 30, 45 and 60 days. The scanning electron microscope (SEM) was used to observe the presence of bacteria at the interface of the root canals.
Results: All six positive controls leaked after day 1, one obturated tooth of the 45-day group (5.26%) leaked, while the others showed no turbidity. The SEM images revealed bacteria accumulated at the interface between root canal wall and root canal fillings as well as in tubular dentine in all groups.
Conclusions: The modified bacterial leakage model showed only 1 obturated tooth leaked in the period from 7 to 60 days. However the SEM revealed that Enterococcus faecalis was present at the interface along the root even when the model did not show turbidity.
